Zusammenfassung: | PROBLEM TO BE SOLVED: To provide an image acquisition method and an image acquisition device that can photograph a biological sample such as a cell in a bright field and obtain an observation image suitable for observation of the biological sample.SOLUTION: A method firstly includes photographing a biological sample at a plurality of wavelength bands with different center wavelengths while radiating illumination light to the biological sample (step S1). The method next includes obtaining a wavelength profile representing change in brightness corresponding to change in a wavelength band for each pixel from a plurality of obtained photographic images (step S2). It then includes classifying each pixel into a plurality of clusters according to the wavelength profile (step S3). It finally includes generating an observation image by allocating a gradation value corresponding to the classified cluster to each pixel (step S4). 【課題】細胞等の生体試料を明視野で撮影し、生体試料の観察に適した観察用画像を取得することができる画像取得方法および画像取得装置を提供する。【解決手段】まず、生体試料に照明光を照射しつつ、中心波長の異なる複数の波長帯域において生体試料を撮影する(ステップS1)。次に、取得した複数の撮影画像について、画素毎に、波長帯域の変化に対する輝度の変化を表す波長プロファイルを得る(ステップS2)。そして、波長プロファイルに応じて、各画素を複数のクラスターに分類する(ステップS3)。その後、各画素に、分類されたクラスターに対応する階調値を割り当てることにより、観察用画像を生成する(ステップS4)。これにより、生体試料の観察に適した観察用画像を生成できる。【選択図】図3 |
