Zusammenfassung: | PROBLEM TO BE SOLVED: To provide an ultrathin membrane for live imaging of suspended cells and a method for producing said membrane.SOLUTION: The porous ultrathin membrane for retaining suspended cells has a thickness of 20 to 200 nm, a through hole diameter of 0.1 to 2 μm and a pitch of 1.5 to 6 μm. The method of producing the porous ultrathin membrane for retaining suspended cells comprises: a step of bringing a mold into contact with the ultrathin membrane side of a composite membrane comprising an ultrathin membrane such as PLA and a sacrificial layer of PVA or the like which is soluble in a liquid medium that does not dissolve the ultra thin membrane and has a thickness of 300 nm or less, heat compressing the composite membrane, and forming through hole in the composite membrane; a step of separating the composite membrane from the mold after forming the through holes in the composite membrane; and a step of separating the porous ultrathin membrane by dissolving the sacrificial layer included in the composite membrane after separating the mold from the composite membrane. The temperature of the mold in the through hole forming step is (the glass transition temperature (°C) of ultra thin film -20°C) to (the glass transition temperature (°C) of ultra thin film +20°C) and the pressure in the through hole forming step is 10 MPa or more.SELECTED DRAWING: Figure 2
【課題】浮遊細胞のライブイメージングの為の超薄膜及び前記膜の製造方法の提供。【解決手段】浮遊系細胞を保定するための、膜厚が20〜200nm、貫通孔の直径が0.1〜2μmでピッチが1.5〜6μmの、多孔質超薄膜。PLA等の超薄膜及び超薄膜が溶解しない液状媒体に可溶で、且つ、膜厚が300nm以下であるPVA等の犠牲層とを含む複合膜に、超薄膜側からモールドを接触させ、複合膜を加熱圧縮して該複合膜に貫通孔を形成する工程、複合膜の貫通孔の形成後、複合膜モールドを分離する工程、及び、複合膜とモールドの分離後、複合膜が含む犠牲層を溶解して多孔質超薄膜を分離する工程、貫通孔形成工程のモールドの温度が、(超薄膜のガラス転移温度(℃)−20℃)〜(超薄膜のガラス転移温度(℃)+20℃)で、貫通孔形成工程の圧力が10MPa以上である、浮遊系細胞保定用多孔質超薄膜の製造方法。【選択図】図2 |
