Zusammenfassung: | PROBLEM TO BE SOLVED: To provide an isomerase for an amino sugar phosphate derived from the genus Sulfolobus for the synthesis of galactosamine-6-phosphate, and a method for producing the same, etc.SOLUTION: According to the present invention, there is provided an isomerase for amino sugar phosphate derived from the genus Sulfolobus. There is also provided a method for producing galactosamine-6-phosphate from glucosamine-6-phosphate by reaction of an isomerase having a specific amino acid sequence at a temperature of 25 to 98°C. Here, the isomerase may be an intramolecular transferase for amino sugar phosphate, and capable of producing galactosamine-6-phosphate from galactosamine-1-phosphate. According to the present invention, there is provided a DNA encoding the enzyme for isomerization. There are also provided a DNA capable of hybridizing with the DNA under stringent conditions and encoding a protein having amino sugar phosphate isomerization activity and an expression vector comprising the DNA. There are further provided a transformant obtained by introducing the expression vector into a host cell and a method for producing galactosamine-6-phosphate by culturing the transformant.SELECTED DRAWING: None
【課題】ガラクトサミン−6−リン酸の合成の為の、スルフォロバス属に由来する、アミノ糖リン酸の異性化酵素、およびその製造方法等の提供。【解決手段】スルフォロバス属に由来する、アミノ糖リン酸の異性化酵素。前記アミノ酸配列を有する異性化酵素は、温度25〜98℃で反応し、グルコサミン−6−リン酸からガラクトサミン−6−リン酸を製造する方法。また、前記異性化酵素は、アミノ糖リン酸の分子内転移酵素であってもよく、ガラクトサミン−1−リン酸からガラクトサミン−6−リン酸を製造できる。前記異性化する酵素をコードするDNA、前記DNAとストリンジェントな条件下でハイブリダイズし、かつアミノ糖リン酸異性化活性を有する蛋白質をコードするDNA、前記DNAを含む発現ベクター、前記発現ベクターを宿主細胞に導入してなる形質転換体及び前記形質転換体を培養して、ガラクトサミン−6−リン酸を製造する方法。【選択図】なし |
