Zusammenfassung: | PROBLEM TO BE SOLVED: To address the problem that although livestock products and aquatic products have relatively low fluorescent noise, agricultural products, especially green and yellow vegetables have a significant problem with fluorescent noise due to the inclusion of pigments, a novel automatic detection method of fluorescent signals has been developed which does not generate noise in an even wider range of foods.SOLUTION: A novel microbial test method which automatically detects fluorescent signals and does not generate noise by detection, by means of detecting a specific color and brightness of a DNA probe-derived fluorescent signal on a membrane filter using a low magnification optical system on proliferating bacteria, using a culture combined in situ hybridization method.SELECTED DRAWING: Figure 1
【課題】畜産物、水産物は、比較的蛍光ノイズが低いが、農産物、取り分け緑黄色野菜は、色素をふくむため、蛍光ノイズの問題が大きい。そこで、より広汎な種類の食品においてノイズの発生しない新たな蛍光シグナル自動検出方法の開発を課題とした。【解決手段】培養併用蛍光インサイチューハイブリダイゼーション法を用いて、増殖された微生物を、低倍率光学系を用いて、メンブレンフィルター上のDNAプローブ由来蛍光シグナルを特定の色と明るさで検出する手段により検出することにより、ノイズの発生しない新たな蛍光シグナルを自動検出する微生物検査方法を提供する。【選択図】図1 |
