Zusammenfassung: | PROBLEM TO BE SOLVED: To provide means of increasing the yield of the refined products of EGF protein that is expressed within a plant body.SOLUTION: The invention provides a manufacturing method of the refined products of the epidermal growth factor that is expressed within a plant body comprising: a crushing step of crushing a plant body containing epidermal growth factor; a centrifugation step of centrifuging a crushing solution and recovering a supernatant; a two-phase partition step of subjecting the recovered supernatant to the aqueous two-phase partition method using the aqueous two-phase system consisting of a polyethyleneglycol phase and a salt phase, and recovering a polyethyleneglycol phase; and a chromatography step of subjecting the polyethyleneglycol phase to an ion exchange chromatography or adsorption chromatography; and comprising no ultrafiltration step.SELECTED DRAWING: Figure 1
【課題】植物体内で発現させたEGFタンパク質の精製品の収率を高めることができる手段を提供すること。【解決手段】上皮細胞増殖因子を含む植物体を破砕する破砕工程;破砕液を遠心分離し、上清を回収する遠心分離工程;回収された上清を、ポリエチレングリコール相及び塩相で構成される水性二相系を用いた水性二相分配法に付し、ポリエチレングリコール相を回収する二相分配工程;及びポリエチレングリコール相をイオン交換クロマトグラフィー又は吸着クロマトグラフィーに付すクロマトグラフィー工程を含み、限外濾過工程を含まないことを特徴とする、植物体内で発現させた上皮細胞増殖因子の精製品の製造方法を提供した。【選択図】図1 |
