Zusammenfassung: | PROBLEM TO BE SOLVED: To provide a measuring method suitable for profiling methylation modification of microRNAs comprehensively and with high accuracy.SOLUTION: Provided is a method for measuring methylation modification sites for microRNAs, including: (1) a step to make a modifier capable of selectively acting on one of methylation candidate sites for ribonucleotides act on the microRNAs; (2) a step to measure fragmentation on the modified microRNAs obtained in the step (1); and (3) a step to compare a detection value which is the mass number of each ribonucleotide, obtained in the step (2) with the mass number of an unmodified corresponding ribonucleotide, and then to determine that the methylation candidate site is methylated when there is no difference between the mass numbers.SELECTED DRAWING: None
【課題】マイクロRNAのメチル化修飾を網羅的にかつ高精度にプロファイリングするのに適した計測方法を提供する。【解決手段】マイクロRNAにおけるメチル化修飾部位を計測する方法であって、(1)リボヌクレオチドのメチル化候補部位の一つに選択的に作用しうる修飾剤をマイクロRNAに作用させる工程;(2)工程(1)で得られた修飾マイクロRNAに対して、質量分析により断片化測定を行う工程;及び(3)工程(2)で得られた、それぞれのリボヌクレオチドについての質量数の検出値を、未修飾の対応するリボヌクレオチドの質量数と比較し、差がないときに前記メチル化候補部位がメチル化されていると判定する工程を含む方法。【選択図】なし |
