Zusammenfassung: | PROBLEM TO BE SOLVED: To provide a method for modifying enzymes to be used in biosynthesis of 1,4-butanediol (1,4-BDO) in E.coli, and a method for screening the modified genes.SOLUTION: Improved microorganism was prepared by introducing and expressing butyraldehyde dehydrogenase and butanol dehydrogenase in E. Coli, where these enzymes are derived from Clostridium saccharoperbutylacetonicum, and their activities in the two terminal reactions in the 1,4-BDO biosynthesis pathway have not been known, that is, in the formation reaction of 4-hydroxybutyraldehyde from 4-hydroxybutyryl-CoA, and in the formation reaction of 1,4-BDO from 4-hydroxybutyraldehyde respectively. As a result, the transformed E.coli was confirmed to have 1,4-BDO productivity. When the activity of butyraldehyde dehydrogenase was strengthened by the directed evolution, the product concentration of 1,4-BDO was increased twice or more. |
