Zusammenfassung:PROBLEM TO BE SOLVED: To adjust the 3'-5' exonuclease activity of a heat-resistant DNA polymerase. SOLUTION: The method for changing the property of the protein comprises introducing a mutation into the amino acid sequences of one or both of portions near to two domains located on the periphery of a groove having a substrate-binding site and/or active center of the groove of the protein having a catalytic activity. Therefore, the distance between both the domains is enlarged, shortend or misaligned each other, thereby changing the size or shape of the groove to change the property of the protein.
