Zusammenfassung:PROBLEM TO BE SOLVED: To provide a simple, sure and specific nonpollution method for point mutation at a low cost, and a new method for genetic manipulation by which practical use can be carried out. SOLUTION: The method for the point mutation of a specific base in nucleic acids comprises irradiating the nucleic acids having a base to be converted such as DNA, RNA or a peptide nucleic acid(PNA) with light in the presence of a DNA containing a photocoupling nucleoside, coupling the base to be converted to the photocoupling nucleoside, chemically modifying the base to be converted, then irradiating the resultant chemically modified base with light and cleaving the resultant substance into the base to be converted and the photocoupling nucleoside. Furthermore, the method is to produce the nucleic acids subjected to the point mutation by the method.
