Zusammenfassung:A method of processing a specimen in a liquid-based cytology preservative comprising formaldehyde, the method comprising combining the specimen with a protease enzyme (proteinase K) and a formaldehyde scavenger (2-imidazolidone), incubating the reaction mixture at an elevated temperature of 85-100 oC, isolating RNA and amplifying the resulting nucleic acid. The incubation temperature may be between 85oC and 95oC. Also claimed is a composition and a kit comprising 2-imidazolidone, proteinase K, EDTA and a pH buffer and an automated system for processing a specimen comprising a pipetting device and a heating element, a reagent vial comprising a formaldehyde scavenger, proteinase K, EDTa and a pH buffer.
