1282405 Piperidine spiro compounds YOSHITOMI PHARMACEUTICAL INDUSTRIES Ltd 6 April 1971 8862/71 Heading C2C Novel piperidine spiro compounds of the general formula wherein Z is either of wherein R is hydrogen, fluorine, chlorine, methyl, methoxy, acetyl or trifluoromethyl; each of R1 and R2 is hydrogen, C 1-4 alkyl or phenyl; R 3 is hydrogen or C 1-4 alkyl; alk is C 1-4 alkylene; X is sulphur, -CH 2 CH 2 -, -CH=CH- or -C(CH 3 ) 2 -; and n is 0 or 1 and salts thereof may be prepared by (i) reacting a compound IV with a compound V (ii) reacting a compound Z-alk-Y with a com pound VII or a salt thereof where Y is halogen or an ester group of an inorganic or organic acid; (iii) when Z is II, reacting Z-M with a compound IX wherein Y is as previously defined and M is hydrogen or an alkali metal; (iv) where Z is III, reacting a protected oxo derivative of a compound Z-alk-Y with a compound VI and removing the protective group by hydrolysis. Pharmaceutical compositions of the compounds I show activity in suppression of spontaneous mobility, lowering body temperature and potentiation of reserpine when administered orally with the usual excipients.
