Zusammenfassung:Producing adherent cells comprises: (a) introducing a suspension of adherent cells into a culture vessel containing microcarriers in a culture medium; (b) amplifying the cells by performing many cell passages in the same culture vessel; and (c) harvesting the cells produced during the last cell passage, optionally after having subjected the cells to enzyme treatment to detach cells from microcarriers. Producing adherent cells comprises: (a) introducing a suspension of adherent cells into a culture vessel containing microcarriers in a culture medium; (b) amplifying the cells by performing many cell passages in the same culture vessel, where each cell passage subsequent to the first cell passage is carried out by using at least part of the cells produced during the previous cell passage after having subjected the cells to enzyme treatment to detach the cells from microcarriers, and introducing culture medium and an increasing amount of microcarriers; and (c) harvesting the cells produced during the last cell passage, optionally after having subjected the cells to enzyme treatment to detach cells from microcarriers. Independent claims are also included for: (1) producing adherent cells comprising thawing a stock of adherent cells, and then subjecting the thawed adherent cells to the process mentioned above; and (2) producing a biological agent from adherent cells comprising: steps (a) and (b) as mentioned above; (c1) treating the cells produced during the last cell passage such that it produces the biological agent, where the treating is carried out in the same culture vessel as the one used to amplify the cells; and (d) harvesting the biological agent. La présente invention a pour objet un procédé de production de cellules adhérentes sur microporteurs selon lequel : a. on introduit des cellules adhérentes dans un récipient de culture qui contient des microporteurs dans un milieu de culture ; b. on amplifie les cellules en effectuant dans ce même récipient de culture plusieurs passages cellulaires successifs en utilisant à chaque fois la population cellulaire provenant du passage cellulaire précédent et en augmentant à chaque fois la quantité de microporteurs que l'on ajoute au milieu de culture; et c. On recueille la population cellulaire provenant du dernier passage cellulaire. Elle concerne également la mise en œuvre de ce procédé pour la production d'agents biologiques, servant notamment à la préparation de vaccins ou de médicaments.
