Zusammenfassung:Genetically engineered microorganism that is not capable of synthesizing a functional protein but can catalyze pathway that allows conversion of 3-hydroxy-3-methylglutaric acid (HMG) to fumaric acid and acetoacetic acid, thus when grows, it accumulates in the culture medium HMG, is new. Independent claims are: (1) a method for obtaining a strain of genetically engineered Pseudomonas putidaso that it is unable to metabolize homogentisic acid, thus accumulating in the culture medium, comprising: (a) amplification by PCR at least two DNA fragments of the genes hmgC and hmgA; (b) binding the amplified fragments and cloned into a vector that also carries a gene for antibiotic resistance resulting in a vector that carries the amplified fragments corresponding to genes hmgC and hmgA; (c) transforming the first vector and selected host microorganism transformed microorganisms resistant to the antibiotic which carried the vector; (d) transferring the vector with the amplified fragments corresponding to genes hmgC and hmgA and strain of Pseudomonas putidawith the help of a third strain of microorganism that facilitates conjugation between genetic material from the first microorganism transformed and acting donor and the genetic material of Pseudomonas putida, which acts as final host microorganism and receiver of the genetic material; and (e) selecting strains of Pseudomonas putidatransformed and unable to metabolize homogentisic acid, by growth on sucrose; and (2) testing device for identifying herbicides, for identifying algaecides, for identifying inhibitors of metabolic pathway enzymes that convert L-Phe, L-Tyr or 3-hydroxyphenylacetate homogentisic acid, comprising genetically modified microorganisms and a precursor homogenistic acid selected from: phenylalanine, tyrosine and 3-hydroxyphenylacetate or its mixtures.
