Zusammenfassung:	Disclosed is a 6'-cyano (CN)-modified locked nucleoside, a nucleotide, and a nucleic acid polymer (NAP), which relates to the field of biotechnology. In the present disclosure, the 6'-CN-modified locked nucleoside has a structure of R configuration or S configuration, and is further modified to obtain the nucleotide and the NAP. Studies have found that the 6'-CN-modified NAP shows better nuclease resistance than unmodified or other modified NAPs, thereby reducing or regulating the interaction between the NAP and intracellular proteins. Therefore, the 6'-CN-modified locked nucleoside, the nucleotide, and the NAP exhibit a significant application value in the field of nucleic acid drugs. Preparation methods of the locked nucleoside, the nucleotide, and the NAP are efficient, stable, and practical, and can be compatible with various substituted or unsubstituted bases.
