Zusammenfassung:	Methods and compositions for monitoring mutation burden, cancer status, vaccine efficacy using cell-free DNA sequencing are disclosed. In some aspects, the method comprises the steps of: a. obtaining or having obtained sequencing data of cell-free DNA (cfDNA) from a sample from the subject, and wherein the sequencing data comprises a target coverage of at least 50% of all polynucleotide regions of interest corresponding to mutations present in an exome of the cancer and wherein the sequenced polynucleotide regions of interest have a mean read depth of at least 1000X.
