Zusammenfassung:	The present invention relates to methods for generating selection marker gene-free recombinant strains. The methods can be applied to organisms having multinucleated cells, such as heterokaryotic cells, of which multinucleated fungal cells are preferred. In the method a multinucleated cell is co-transformed with at least two separate nucleic acid constructs, one construct comprising a selection marker and the other construct comprising a gene of interest. The method then employs a combination of selection, screening and segregation of multinucleated heterokaryotic cells into homokaryotic cells to select marker gene-free recombinant strains comprising the gene of interest.
