The systems and compositions can include levulinic acid in an amount to enhance transdermal permeation of estradiol and a pharmaceutically acceptable carrier. The systems and compositions can optionally include a solubilizing agent and additional permeation enhancers. Pharmaceutically acceptable carriers can include adhesives, particularly acrylate copolymers, gels, creams, ointments, solutions, suspensions, and pastes.
