Zusammenfassung: | Detecting a prion protein PrP in a biological sample of animal or human origin consists of using a molecule with at least one positive charge and/or at least one osidic link and a ligand other than the protein ligand. The molecule is added to the biological sample to cause precipitation of the PrP before adding the ligand other than the protein ligand, and preferably a macrocyclic ligand, while a supplementary stage consists of adding proteinase K to the sample to digest a cellular prion protein (PrPc). The procedure consists of adding the proteinase K to the biological sample, adding the positively-charged molecule to the mixture, followed by the ligand other than the protein ligand, and revealing the resistant pathogenic protein (PrPres). |
