Zusammenfassung:	The present invention relates to methods for the PCR-based detection of nucleic acids from Mycobacterium avium ssp. paratuberculosis (Mycobacterium paratuberculosis), in biological samples such as milk. The oligonucleotide primers to be used in the method of the invention are designed so as to improve the specificity of detection of Mycobacterium avium ssp. paratuberculosis by avoiding detection of related environmental Mycobacteria. The sensitivity of detection is improved by using an optimized procedure for extraction of DNA from milk and by using a nested PCR method.
