Zusammenfassung:A labeled primer (for example for the polymerase chain reaction) is described, which primer is labeled at the two ends of the oligonucleotide strand with a reporter dye molecule and a quencher molecule, and in which labeled primer at least the last base at the 3' end is not complementary to the DNA sequence to be amplified. A process for detecting a DNA sequence (by means of the polymerase chain reaction) using the labeled primer is also described, which process uses, for the amplification, one or more thermostable DNA polymerases, one of which also possesses proof-reading properties and releases the unpaired bases of the labeled primer together with the quencher molecule which is attached thereto, resulting in an increase in the fluorescence at the wavelength of the reporter dye and thereby indicating the formation of the DNA sequence which is labeled with this fluorescent dye.
