Zusammenfassung:The invention is directed to improved methods for modification of target nucleic acids in a cell, wherein a fusion nucleic acid (fuNA) molecule, comprising a guide nucleic acid (gNA) molecule covalently linked to at least one donor nucleic acid (doNA) molecule, and introducing said fuNA molecule into one or more cells comprising the target NA molecule, and introducing into one or more cells a site directed nucleic acid modifying polypeptide, e.g. Cas9. Homology arms in the donor nucleic acid which may be RNA, DNA or PNA, have at least 15 nucleotides 100 % complementary to 15 nucleotides in the target. Also claimed is a fuNA molecule which consists of RNA.
