Zusammenfassung:	The present invention relates to biology field, the connection and determination method of oligonucleotide library small sequence with fixed sequences on its two ends. Once sequencing can obtain 8-20 library small sequences, and this means 8-20 times raised sequencing effect and 8-20 times lowered cost. One special primer, bridging primer, is designed for PCR amplification and the amplified product is cloned to relevant vector before sequencing. The method is fast and economical in determining oligonucleotide library small sequence with fixed sequences on its two ends. The present invention may be applied in describing oligonucleotide library sequence combined to large biological molecules, and has application value in theory research and human disease treating medicine research.
