Zusammenfassung: | The method for controlling space orientation of immobilized enzyme includes the following steps: utilizing gene manipulation to constitute and express enzyme-joining peptide-streptoconjugated peptidefusion protein; preparing fusion protein pure product; ultrafiltration and concentration, measuring enzyme kinetics, utilizing specificity combination of streptoconjugated peptide and streptavidin tomake enzyme molecule orientationally immobilized on the solid phase carrier coated with streptavidin, and its active centre is toward reaction solution. The fusion protein is a "anchor-strand" molecule system, in which the streptoconjugated peptide is the "anchor" for controlling immobilizing direction of enzyme moleule, and the joining peptide is the "strand" for increasing molecule distance between enzyme and streptoconjugated peptide, so that it not only can keep the flexibility of their respective space movement, but also keduce steric hindrance of their combination reaction so as to greatly raise its orientation immobilizing efficiency. It is suitable for making high-quality biological devices of biosensor and biochip, etc. |
