The method comprises the following steps: S1, selecting a 3K or 9K bacterial culture medium to culture to-be-rejuvenated bacteria to a plateau phase to obtain a rejuvenated flora, the to-be-rejuvenated bacteria comprising mesophilic bacteria RetechV or moderate thermophilic bacteria RetechM III; the rejuvenation flora is domesticated for multiple rounds under the condition that the ore pulp concentration is 2% or 5%, the number of bacteria in the rejuvenation flora reaches 108/ml or above, and the potential reaches 600 mv or above; s2, after the fineness of the lead concentrate is ground to the target fineness, the ore pulp concentration is set to be 2%-10%, the initial pH value is set to be 1.2-1.8, and to-be-leached ore pulp is obtained; and S3, domesticated rejuvenation flora is added into the ore pulp to be leached, the leaching time is 7-30 days, and the
