The amino acid sequence of patchouli alcohol synthase PcV1 is as shown in SEQ ID NO. 1; according to the patchouli alcohol synthase PcV1 mutant, leucine at the 297th site of an amino acid sequence as shown in SEQ ID NO.1 is mutated into isoleucine, as shown in SEQ ID NO.2; compared with a wild expression strain PcV1, the catalytic activity of the alpha-butene of the engineering strain disclosed by the invention is obviously improved, the yield of patchouli alcohol of the PcV1 is 32.96 mg/L, and the yield of the alpha-butene is 27.04 mg/L; according to the L297I mutant, the yield of the patchouli alcohol is 1.31 mg/L, the yield of the alpha-butene is 78.28 mg/L, and conditions are created for industrial production of the alpha-butene.
