The construction method comprises the following steps: replacing a tyR gene with a Ptrc-aroGS180F gene, replacing a yhgX gene with a Ptrc-serAH344A/N364A gene, and replacing a trpE gene with a Ptrc-trpES40F gene. And the tnaA gene is replaced by the gene luz15 gene. The yield of the 5-hydroxytryptophan of the recombinant MG1655-strain prepared by using the construction method disclosed by the invention can reach 105 mg/L. 本发明提供一种合成5-羟基色氨酸大肠杆菌MG1655菌株的构建方法,所述构建方法包括如下步骤:将Ptrc-aroGS180F基因替换tyR基因、Ptrc-serAH344A/N364A基因替换yhgX基因、Ptrc-trpES40F基因替换trpE基因。并将基因luz15基因替换tnaA基因。使用本发明所述构建方法制得的重组MG1655-菌株的5-羟基色氨酸产量可达105mg/L。
