The method comprises the following steps: constructing an ultrasonic time difference method flow model; calibrating optimal solution parameters in the ultrasonic flow model by adopting a cuckoo multi-objective optimization algorithm; the optimal solution parameters are input into the constructed ultrasonic time-difference method flow model, and an optimal ultrasonic time-difference method flow model is obtained; and inputting the measured value of the local flow of each layer of the river flow cross section into the optimal ultrasonic time-difference method flow model, and outputting the optimal calculation value of the average flow of the river flow cross section.
