Zusammenfassung:The invention belongs to the technical field of biological materials, and particularly discloses a transformation method for improving affinity of RHD1 and a linear ubiquitin chain and retaining specificity of the RHD1. Amino acid of a binding interface of the RHD1 and the linear ubiquitin chain is analyzed by utilizing bioinformatics software based on RHD1 (1-200aa) and linear double ubiquitin structure data; the method comprises the following steps of: selecting important amino acid sites for point mutation or combinatorial mutation, determining whether the mutation improves the affinity of combining with a linear ubiquitin chain or not through enzyme-linked immunosorbent assay (ELISA), developing the RHD1 into a protein ligand capable of specifically identifying and efficiently combining with the linear ubiquitin chain, and providing a powerful tool for specifically identifying the linear ubiquitin chain and researching the function of the linear ubiquitin chain. 本发明属于生物材料技术领域,具体公开了一种提高RHD1与线性泛素链亲和力并保留其特异性
