Zusammenfassung: | The invention discloses an SERS (Surface Enhanced Raman Scattering) sensor for DNA (Deoxyribose Nucleic Acid) methylation detection as well as a preparation method and application of the SERS sensor. The SERS sensor comprises a pentamethylcytosine antibody modified nanogold-coated magnetic bead as a capture substrate, and a specific gene sequence modified on gold-silver core-shell nanoparticles embedded with different internal standard molecules as a detection probe; the invention also discloses simultaneous detection of methylation levels of related genes in a lung cancer sample by combining the sensor with a surface enhanced Raman spectroscopy technology, and the method can realize methylation detection without depending on any complex sample pretreatment and nucleic acid amplification technology, and has the advantages of cost saving, high universality, high sensitivity and the like. The method has huge potential in disease diagnosis and clinical medical research.
