Zusammenfassung: | The invention discloses a dual-luciferase reporter gene vector based on a human LDHA gene promoter region as well as a construction method and application of the dual-luciferase reporter gene vector. The dual-luciferase reporter gene vector is marked as pGL3-LDHA-promoter with a nucleotide sequence as shown in SEQ ID NO.2. The dual-luciferase reporter gene vector is constructed by utilizing an efficient enzyme and a homologous recombination principle. BgL II and HindIII are utilized to complete cloning and recombination of an LDHA gene promoter target fragment and a pGL3-Basic linearized vector, and a double enzyme digestion method can greatly reduce self-connection of the vector and improve the positive rate of the recombinant vector. The pGL3-LDHA-promoter can be used for realizing sensitive identification and accurate detection on the expression quantity of the LDHA promoter.
