Zusammenfassung:The invention discloses a method for exploring peripheral proteins of a leading chain and a following chain based on an APEX2 proximity marker technology, which comprises the following steps of: constructing a fusion protein carrier containing an EFI alpha promoter sequence, APEX2 and V5 by selecting a key protein related to DNA (Deoxyribonucleic Acid) copying the leading chain and the following chain as a target, and transforming the fusion protein carrier into a host cell line to form a fusion cell line; the method comprises the following steps: performing biotin labeling on adjacent proteins through biotin-phenol and hydrogen peroxide treatment, then extracting and purifying biotinylated proteins, performing quality verification, and finally performing mass spectrum and chromatography on-machine analysis.
