The invention aims to construct a pGhRBE3-pGh alpha GloA-GhPGF expression vector by adopting a cotton endogenous pGh alpha GloA promoter to drive crRNAs of a GhPGF gene, so that a cotton CRISPR/Cas12a gene editing system is optimized, the editing efficiency of the gene editing system for a single crRNA is up to 100%, and the efficiency of the gene editing system for simultaneously editing two crRNAs can reach 66.7%, thereby providing a new scheme for cotton genome editing.
