The invention specifically discloses amino acid mutation of the Bst DNA polymerase mutant on an amino acid sequence of a wild type Bst DNA polymerase large fragment, wherein the amino acid mutation comprises H31I, C91K, S258D and T307P; the amino acid sequence of the wild type Bst DNA polymerase large fragment is as shown in SEQ ID NO: 1. Compared with a wild type Bst DNA polymerase, the Bst DNA polymerase has higher thermal stability, the yield of reaction products is higher, and the Bst DNA polymerase has a good application prospect in isothermal amplification.
