Zusammenfassung: | The invention discloses a method for preparing a brain-like organ from in-vivo source haploid cells, which comprises the following steps: step 1, obtaining a reconstructed embryo from BCL2 overexpressed mouse haploid embryonic stem cells by an embryo injection method, transplanting the reconstructed embryo back to a fallopian tube of a 0.5-day pseudopregnant female mouse, dissecting when the embryo develops to E8.5 days, and taking out the embryo; separating body segment tissues; 2, culturing the separated somatic segment tissues in a neural stem cell culture medium, enriching haploid cells through flow sorting, and establishing a mouse haploid neural stem cell line; 3, culturing the obtained mouse haploid neural stem cell line in a brain-like culture solution, and performing growth and space folding to obtain a mouse brain-like organ derived from haploid neural stem cells; and 4, identifying by adopting immunofluorescence cells and molecular biology.
