The purification method comprises the following steps: A, balancing an affinity chromatography column by using an equilibrium buffer solution; b, loading the cell fermentation supernatant containing the anti-canine IL-31 monoclonal antibody to an affinity chromatography column; c, balancing the affinity chromatography column by using the equilibrium buffer solution again until the cell fermentation supernate completely flows through the affinity chromatography column; d, leaching the affinity chromatography column by using a leaching buffer solution; e, balancing the affinity chromatography column by using the equilibrium buffer solution again; and F, eluting the affinity chromatography column by using an eluent, and collecting the eluent containing an elution peak of the anti-canine IL-31 monoclonal antibody, so as to obtain a purified solution of the anti-canine IL-31 monoclonal antibody.
