The invention also discloses a preparation method of the U6 promoter of idesia polycarpa. Four U6 promoters cloned from idesia polycarpa are connected to a pGreenII0800 vector containing an LUC reporter gene, tobacco leaves are instantaneously converted through agrobacterium, it is found that the cloned four IpU6 promoters all have activity, the transcriptional activity of IpU6-2 and IpU6-1 is higher than that of other two IpU6 promoters, and the transcriptional activity of IpU6-2 and IpU6-1 is higher than that of a control OsU6a, the transcriptional activity of IpU6-2 and IpU6-1 is higher than that of a control OsU6a, and the transcriptional activity of IpU6-2 and IpU6-1.
