The invention provides an application of a substance for detecting polymorphism or genotypes of three KASP markers, namely Wx-In1-KASP, Wx-Ex10-KASP and SSIIa-KASP, in identification or auxiliary identification of the quality of rice. The KASP marker is developed according to G or T located at a first intron + 1 of Wx, C or T located at a tenth exon + 115 of Wx, and C or T located at the beginning of ATG of an SSIIa transcript + 2341. The genetic distance between the KASP markers Wx-In1-KASP, Wx-Ex10-KASP and SSIIa-KASP and the target genes Wx and SSIIa is 0 cM, and the molecular markers which are most closely linked with the three sites of the two target genes are obtained.
