Zusammenfassung: | The invention discloses application of ZmGCH2 protein and a coding gene thereof in regulating folic acid content of plant seeds. The method comprises the following steps: firstly, carrying out conjoint analysis and comparison on transcriptomes and proteomes of three fresh waxy corn inbred lines with high folic acid content, middle folic acid content and low folic acid content, and identifying and cloning to obtain a ZmGCH2 gene related to the folic acid content of a plant; further, by constructing a ZmGCH2 mutant plant and an overexpression plant and measuring the content of folic acid in grains, the content of 5-formyltetrahydrofolic acid and the content of 5-methyltetrahydrofolic acid in the grains of the ZmGCH2 overexpression plant are respectively 4.3 times and 2.8 times that of a wild type; the folic acid content in ZmGCH2 mutant plant grains is slightly lower than that of a wild type and does not reach a significant level.
