According to the invention, source screening and rational transformation of the phycocyanobilin ferredoxin reductase (PcyA) are realized, and the problem of low yield of phycocyanobilin in the prior art is solved. A key rate-limiting enzyme which is most suitable for biliverdin and phycocyanobilin escherichia coli synthesis is screened from PcyA from five different algae sources, and rational transformation is carried out on the basis, so that high-efficiency synthesis of shake flask level phycocyanobilin in escherichia coli can be realized. At the shake flask level, the yield of the obtained phycocyanobilin can reach 24.80 mg/L, and the result lays a foundation for application of the phycocyanobilin in the fields of food, medicine and cosmetics.
