Zusammenfassung: | The invention discloses a multi-sgRNA (small guide ribonucleic acid) mediated EXPERTplus pilot gene editing system and application of the multi-sgRNA mediated EXPERTplus pilot gene editing system. In order to improve the editing efficiency of the EXPERT system, on the basis of the EXPERT system, a plurality of sgRNAs are additionally arranged between the orii-sgRNA and the expegRNA. The result shows that the EXPERTplus system can further improve the editing efficiency. Compared with a PE system, the EXPERTplus system has the advantages that the editable range is wider, the accurate replacement of 88bp can be realized, and the editing efficiency is higher. Besides, the developed EXPERTplus system only generates a notch on one chain and does not generate DNA double-chain breakage, so that compared with a PE3 system and a twin PE system which may generate DNA double-chain breakage, the EXPERTplus system can generate lower indels, and is safer.
