The nucleotide sequence of the primer comprises the sequences as shown in SEQ ID NO. 1 to SEQ ID NO. 2; the nucleic acid sequence of the probe comprises a sequence as shown in SEQ ID NO. 3. Specific primers and probes are designed for CAR cells containing CD8 alpha transmembrane region-41 BB costimulatory molecules, a fluorescent quantitative PCR method for detecting the copy number of CAR genes is developed, then the CAR cells are analyzed, and the primer and probe combination is proved to have wide applicability through multi-angle verification analysis and limitation of detection standards.
