According to the method, polypeptide containing a fluorophore and a fluorescence quenching group (FRET effect) is used as a substrate, the FRET effect can enable the fluorescence effect of the substrate polypeptide to disappear, and the FRER effect is inhibited and the fluorescence intensity is increased under the action of cutting the substrate polypeptide by Mpro.
