Zusammenfassung:Provided is a method for producing a purine-based substance such as inosine. In the present invention, a purine-based substance is produced by culturing a bacterium that has a purine-based substance-producing ability and belongs to the family Enterobacteriaceae in a culture medium and collecting a purine-based substance from the culture medium and/or a cell of the bacterium, the bacteria are modified so as to have one or more modifications selected from the group consisting of modifications (A) to (C): (A) a modification in which the activity of the PAJ3461 protein is reduced; (B) a modification in which the activity of the PAJ3462 protein is reduced; and (C) a modification in which the activity of the PAJ3463 protein is reduced. 本发明提供一种肌苷等嘌呤类物质的制造方法。本发明中，将具有嘌呤类物质生产能力并且属于肠杆菌科的细菌在培养基中培养，并从该培养基和/或该细菌的菌体中采集嘌呤类物质，从而制造嘌呤类物质，其中，所述细菌被修饰为具有选自下述(A)～(C)的修饰中的一种或一种以上的修饰：(A)PAJ_3461蛋白的活性降低的修饰；(B)PAJ_3462蛋白的活性降低的修饰；(C)PAJ_3463蛋白的活性降低的修饰。
