The preparation method comprises the following steps: taking methyl glyoxylate as a raw material, carrying out coupling reaction on the methyl glyoxylate to generate an intermediate II, cyclizing the intermediate II to generate an intermediate III, and carrying out in-situ methylation on the intermediate III to obtain the 2, 5-dimethyl-4-hydroxy-3 (2H)-furanone. The structural formulas of the intermediate II and the intermediate III are as follows: # imgabs0 # imgabs1 #. After the obtained intermediate II is placed at 35 DEG C for 60 days, the gas phase content of the intermediate II is basically not changed, so that the stability of the intermediate II is superior to that of an intermediate synthesized by taking pyruvic aldehyde as a raw material, and the yield (94%) of the intermediate II is far higher than that of the intermediate synthesized by taking the p
