The method specifically comprises the following steps: carrying out ethanol precipitation and rinsing on Trizol homogenate of an interlayer/organic layer containing DNA, and dissolving with NaOH to obtain a neutralized solution; adding protease K into the obtained solution, and performing water bath digestion to obtain a reaction solution; adding a DNA binding solution into the reaction solution, then adding isopropanol, uniformly mixing, adding a purification column, and centrifuging; adding DNA rinsing liquid into the purification column, and centrifuging; adding water into the purification column for elution, standing and centrifuging to obtain a DNA sample.
