According to the method disclosed by the invention, plant embryos with high grease and high starch content are subjected to splitting decomposition and solid-liquid separation, so that plant cell nucleuses are fully released; the cell nucleus suspension is subjected to Percoll solution density gradient centrifugation, so that more complete cell nucleuses can be obtained through separation.
