The invention provides a polynucleotide sequence which is optimized by synonymous codon preference and is used for coding uricase, and the sequence has high expression efficiency in escherichia coli; the invention also provides a carrier adapted to the polynucleotide sequence, the high-activity uricase is prepared through the synergistic cooperation of the carrier and the polynucleotide sequence, and the specific activity is as high as 32.2 U/mg; in addition, the invention further provides a method for preparing the recombinant uricase, the yield of the uricase is increased, the content of soluble protein in the obtained product is extremely high, the product is easy to purify, post-treatment steps are simplified, and the production cost is saved.
