The invention belongs to the field of adsorbent preparation, and particularly relates to a copper-based adsorbent, a preparation method thereof and application of the copper-based adsorbent in removal of carbon monoxide in hydrogen. The preparation method comprises the following steps: 1) preparing a copper salt precursor solution, and adding an organic compound dispersant into the copper salt precursor solution; wherein the organic compound dispersing agent is at least one of ethylene glycol, sec-butyl alcohol and citric acid; and 2) dipping a Y-type molecular sieve carrier in the precursor solution prepared in the step 1), drying and roasting to prepare the copper-based adsorbent of the Y-type molecular sieve.
