The allelic variation of TCCC or TCC, TC and T at the-1303 site affects the expression quantity of Ctb1 in the germplasm resource population, so that the population can be divided into a strong low-temperature-resistant type (HapI), a medium low-temperature-resistant type (Hap II) and a low-temperature sensitive type (HapIII). An isogenic line (Ctb1HapI) constructed by taking the Huazhan as a receptor parent (Ctb1HapIII) is verified, and the isogenic line (Ctb1HapI) shows higher low temperature resistance than the Huazhan, so that the feasibility of taking the sequence of the Ctb1 promoter region-1303 position as the molecular marker is realized.
