The invention relates to a cRGD (arginine-glycine-aspartic acid) modified xanthine and a derivative nano-drug thereof and a preparation method of the cRGD modified xanthine and the derivative nano-drug thereof, and designs and synthesizes a nano-preparation which is obtained by efficiently modifying a targeting group cRGD on the surfaces of xanthine, 8-epi-xanthine, xanthinosin and XTT (1 beta-hydroxyl-5alpha-chloro-8-epi-xanthine) and encapsulating the surfaces of the xanthine, the 8-epi-xanthine, the xanthinosin and the XTT into a nano-carrier. Therefore, the targeting property of the natural medicine is enhanced, and the action selectivity and bioavailability of the medicine are improved.
